Anti-Diabetic Activity of Heuksoojeongchal Bran Prethanol Extract in HFD/STZ-Induced Diabetic Mice

Abstract
Heuksoojeongchal was developed as a new black barley variety in the National Institute of Crop Science, RDA. We had previously discovered that Heuksoojeongchal bran prethanol extract (HSB) exhibits ¥á-glucosidase inhibition activity. Therefore, this study sought to evaluate the anti-diabetic effects of HSB and its underlying mechanism using high-fat diet (HFD) and streptozotocin (STZ)-induced diabetic mice. HSB (200 mg/kg) was orally administered to the diabetic mice for 6 weeks. HSB significantly reduced fasting blood glucose compared to the control group. Insulin immunohistochemistry staining revealed that the insulin expression in the ¥â-cells of the islets of Langerhans in the HSB group was greater than that of the control. HSB increased hepatic glycogen content and glucokinase mRNA expression and decreased gluconeogenic genes (G6Pase and PEPCK). HSB significantly lowered serum and hepatic lipid contents as well as hepatic lipid droplets in diabetic mice, which are attributable to the down-regulated lipogenic gene (PPAR¥ã) and up-regulated lipid oxidation genes (PGC1¥á, ACSL1, and CPT1¥á) in the liver. Furthermore, HSB significantly down-regulated hepatic inflammatory genes (NF¥êB, TNF¥á, and IL-6) expression in diabetic mice. Thus, HSB improved HFD/STZ-induced diabetes by partly modulating the glucose, lipid, and inflammation metabolism-related gene expression.
